目的探讨中药有效成分黄芩苷对白念珠菌凋亡的影响。方法采用Hoechst33258染色荧光显微镜检测白念珠菌细胞凋亡的形态;Rh123染色流式细胞仪检测细胞线粒体膜电位(mitochondrial membrane potential,MMP)变化;DHR染色流式细胞仪检测细胞内活性氧(reactive oxygen species,ROS)水平。结果 1 000,100μmol/L的黄芩苷能诱导白念珠菌细胞核固缩与碎裂,并降低白念珠菌MMP;1 000,100,10μmol/L的黄芩苷均能提高细胞内ROS水平。结论黄芩苷可通过线粒体途径诱导白念珠菌凋亡。
AIM To investigate the apoptotic effect of baicalin on Candida albicans. METHODS Fluorescence microscope was used to observe the morphological change of apoptotic Candida albicans cells by Hoechst 33258 staining. Flow cytometry was used to analyze the mitochondrial membrane potential (MMP) of Candida albi- cans cells by rhodamine 123 staining and to measure the levels of reactive oxygen species (ROS) by DHR staining. RESULTS 1 000, 100 ~mol/L of baicalin could induce nuclear condensation and fragmentation and decrease MMP of Candida albicans, and 1 000, 100, 10 umol/L of baicalin could increase intracellular ROS accumulation of Candida albicans. CONCLUSION The study confirms that baicalin could induce apoptosis of Candida albicans by mitochondrial pathway.
