目的观察白介素(interleukin,IL)-10、肿瘤坏死因子(tumor necrosis factor,TNF)-α在脓毒症小鼠小肠黏膜中的表达及其意义。方法 40只BALB/c小鼠随机分为模型组(脓毒症模型)和正常对照组,采用改良升结肠支架置入腹膜炎(colon ascendens stent peritonitis,CASP)脓毒症小鼠模型,通过ELISA法检测小肠组织匀浆IL-10、TNF-α的表达水平。结果模型组小鼠外周血TNF-α和IL-10迅速升高,TNF-α高峰的出现早于IL-10,尤其前6h TNF-α上升明显;IL-10水平呈现高表达,持续时间长于TNF-α,直到72h后,IL-10的水平仍然较高。结论脓毒症小鼠动物模型建立后的前12h,是炎症反应的最重阶段。
Objective The aim of our study was to investigate the role of IL-10 and TNF-α in a murine sepsis model. Methods 40 BALB/c rats were randomly divided in to experimental group,which the colon ascendens stent peritonitis model(CASP)was applied,and control group. Then,the expression levels of IL-10 and TNF-α in small intestine tissues were measured by ELISA. Results Comparing with control group,significantly increased IL-10 and TNF-α levels were observed in the CASP group.Meanwhile,TNF-α reached the maximum level much earlier than IL-10,especially in the first 6 hrs. We also demonstrated that the duration of IL-10 peak level was longer than TNF-α,which significantly higher after 72 hrs. Conclusions The severest inflammatory response period was the first 12 hrs after CASP were applied.
