目的 探讨Fos蛋白对前原速激肽A(PPTA)基因表达的调节作 用。方法 免疫组织化学和原位杂交组织化学技术。结果 足底皮下注入蜜蜂毒引起持续、单向性的痛行为反应,导致Fos蛋白在刺激侧脊髓 背角神经元的表达以及PPTA mRNA阳性神经元的数量增加。椎管内给予c-fos 反意寡核苷酸(ASO)明显抑制蜜 蜂毒诱导的痛反应和减少脊髓背角Fos蛋白和PPTA mRNA阳性神经元的数量。椎管内给予盐水 和c-fos正义寡核苷酸(SO)则无影响。结论 Fos可 能参与伤害性刺激诱 导的脊髓背角神经元PPTA mRNA的表达上调,在外周伤害性信息的传递中可能起着重要的作用。
Objective To investigate the roles of Fos protei n in the regulation of prepro tachykinin A (PPTA)gene expression.Metho d s Intraplantar injection of bee venom (BV) was used as a pain model.Immu n ohistchemistry and in situ hybridization were performed to obser ve the effects of intrathecal pretreatment of c-fos antisense oligodeoxynucleotid e (ASO) to the expr ession of Fos protein and PPTA mRNA expression in the spinal dorsal horn.Results Administration of BV into the plantar surface produced a ton i c, monophasic nociceptive response,induced Fos expression in the neurons of th e ipsilateral spinal dorsal horn and increased the number of PPTA mRNA Positive ne urons.Intrathecal pretreatment with c-fos ASO not only significantl y inhibited t he BV-induced nociceptive response but also reduced the number of Fos and PPTA mRNA positive neurons in the spinal dorsal horn.Conclusions F os might play important roles in the processing of noxious information and take pa rt in the noxious stimulation-induced upregulation of PPTA mRNA expression in t he neurons of spinal dorsal horn.
