使用海洋枝顶孢霉(Acremoniumsp.BH0531)活性代谢物处理松材线虫,通过线虫的形态、运动和酶活的变化对BH0531杀线虫机理进行初步探讨。结果表明,经BH0531代谢物处理后的线虫虫体严重扭曲,扭动频率显著提升,并随时间推移而趋缓,最终呈C形、新月形或直形死亡。酶活性测试表明,该菌代谢物对松材线虫体内的乙酰胆碱酯酶AChE具较强抑制作用,酶比活力由0.417 U/mg降至0.089 U/mg;ATP酶活力先升高后下降,处理2h酶比活力由0.447U/mg升至0.501U/mg,随着时间的持续又急剧下降至0.18 U/mg左右。这两种酶活力的变化情况与线虫处理后运动频率变化的观察结果相一致。
Marine-derived Acremonium sp. BH0531 was employed to detect its influence on the morphology, athletic ability and enzymatic activity of Bursaphelenchus xylophilus. Results showed that, when treated with the BH0531 metabolites, the morphology and athletic ability of the nematodes were significantly accelerated, as time went by, decrease was observed, finally the nematodes dead in shapes of C, new-moon or straight. Enzyme activity test showed that the metabolites had strong inhibitory effect on acetylcholin esterase AChE, and the enzyme activity decreased obviously from0.417 U/mg to 0.089 U/mg, while the ATPase increased from 0.447 U/mg to 0.501 U/mg at beginning and then decreased to low about 0.18 U/mg later. The variation trend of the two enzyme activities were correspond to the movement rate of nematodes.
