对分布于我国海南东部海域的波纹裸胸鳝群体(n=21)的mtDNA D-loop区987 bp全序列进行了分析。结果表明:21条波纹裸胸鳝D-loop区序列中,A+T平均含量为62.5%;经比对,共检测到177个核苷酸多态位点,约占核苷酸总数的18.7%;D-loop全序列突变类型有5种,即转换158、颠换18、插入2、缺失1及转换与颠换共存1,它们分别占核苷酸多态位点的89.3%、10.1%、0.3%、0.15%及0.15%。由此构建了中国海南沿海21条波纹裸胸鳝的NJ分子系统树,聚类分析表明:所测波纹裸胸鳝的mtDNA D-loop序列表现为2个单倍型组,从而可以看出这21条波纹裸胸鳝可能有2个母系起源,从系统树上来看,它们之间并没有明显的地域差异。
In this study,987 bp sequences of mtDNA D-loop region,from 21 UndμLatus Gymnothorax in Hainan coast in China,were analyzed.The results showed that: among all the D-loop sequences in the 21 U.Gymnothorax,the content of A+T reached 62.5% on average;there were 177 nucleotide polymorphisms detected by comparison,accounting for 18.7% in the total number of nucleotide;there were 5 types of mutations in D-loop sequences that 158 transitions,18 transversions,2 inserts,1 missing and 1 coexistence of transition and transversion,which accounted for nucleotide polymorphisms 89.3%,10.1%,0.3%,0.15% and 0.15%,respectively.Thus the NJ phylogenetic trees of this 21 U.Gymnothorax in Hainan's coast were constructed.The cluster analysis showed that: all the measured sequences assumed to be two haplotypes.So it could be inferred that the 21 U.Gymnothorax may originated from two maternal ancestors and there were no obvious geographical differences between them seen from genealogical tree.
