目的探讨转基因增强绿色荧光蛋白小鼠血小板黏附和聚集功能的变化。方法选用12~14周龄转基因增强绿色荧光蛋白小鼠和C57BL/6J小鼠,戊巴比妥钠麻醉,心脏取血,肝素抗凝,全自动血细胞分析仪测定血小板参数,灌流小室法测定血小板在胶原面的黏附变化,比浊法测定不同浓度二磷酸腺苷诱导下的血小板最大聚集率。结果与转基因增强绿色荧光蛋白雄性小鼠比较,雌性小鼠的血小板数、血小板平均体积、血小板压积、黏附率和最大聚集率(二磷酸腺苷3、10和30μmol)下降(P〈0.05);与C57BL/6J雌性小鼠比较,转基因增强绿色荧光蛋白小鼠血小板数降低(P〈0.05)。结论转基因增强绿色荧光蛋白雌雄小鼠之间血小板数、血小板平均体积、血小板压积和黏附聚集功能有差异,转基因增强绿色荧光蛋白小鼠与C57BL/6J雌性小鼠之间血小板数也有差异。
Objective To measure the changes of platelets adhesion and aggregation function in transgenic enhanced green fluorescent protein(EGFP) mice. Methods Transgenic EGFP and C57BL/6J mice(12-16 weeks, both male and female) were used for the present study. The mice were anesthetized with Pentobarbital sodium and blood was taken from the heart anticoagulated with Heparin. Platelet parameters were measured using an automated blood cell analyzer. Platelet adhesion on collagen surface was evaluated using a welldefined perfusion chamber. The maximum aggregation rate of platelets induced by ADP was determined by turbidimetry. Results In transgenic EGFP mice, platelet count, mean platelet volume(MPV), plateletcrit(PCT),adhesion rate and the maximum aggregation rate(ADP final concentration was 3 μmol, 10 μmol and 30 μmol respectively) of the female mice were decreased compared with those of the male ones(P〈0.05). In the female mice, platelet count of the transgenic EGFP mice were decreased compared with that of the C57BL/6J mice(P〈0.05). Conclusions There are differences in platelet count, MPV, PCT, adhesion rate and the maximum aggregation rate between male and female transgenic EGFP mice. There is also difference in platelet count between female transgenic EGFP mice and C57BL/6J mice.
