目的:通过研究自发性高血压大鼠(SHR)外周血淋巴细胞电压依赖性钾通道1.3(Kv1.3)mRNA表达的变化,为SHR淋巴细胞激活提供证据.方法:使用密度离心法采集10只SHR和10只正常Wistar大鼠外周血淋巴细胞,运用实时荧光定量聚合酶链式反应(PCR)技术检测外周血淋巴细胞Kv1.3 mRNA表达水平.结果:(1)与对照组相比,SHR组Kv1.3 mRNA表达水平明显升高[(0.0023±0.0005)Kv1.3/β-actin mRNA∶(0.013±0.017)Kv1.3/β-actin mRNA,P<0.05];(2) SHR收缩压和淋巴细胞Kv1.3mRNA表达呈正相关(r=0.830,P=0.003).结论:SHR淋巴细胞Kv1.3 mRNA表达增高,提示钾通道可能在高血压病淋巴细胞激活中起着重要的调控作用,推测淋巴细胞钾通道增多可能与高血压的发生、发展密切相关.
Objective: To determine express of Kv1. 3 mRNA in lymphocyte derived from spontaneously hypertensive rat (SHR), which aimed to identify the relationship between lymphocyte Kv1. 3 and hypertension. Methods: Peripheral blood were collected from SHRs (10 cases) and normotensive Wistar rats (10 cases). Peripheral lymphocytes were separated from heparinized whole blood by standard Ficoll-Hypaque density gradient centrifugation. Real-time PCR was used to detect the express of Kv1. 3 mRNA in lymphocyte. Results: (1) The expression levels of Kv 1.3 mRNA in lymphocytes from SHRs significantly increased compared with that of Wistar rats [ (0. 0023±0. 0005) Kvl. 3/β-actin mRNA : (0. 013±0. 017) Kv1. 3/β-actin mRNA, P〈0. 051; (2) Linear correlation existed be- tween systolic blood pressure and expression of Kvl. 3 mRNA in lymphocytes from SHR (r=0. 830, P=0. 003). Conclusion: The lymphoeytes Kv1. 3 mRNA expression level up regulate in SHR, it suggests Kv1. 3 contribute to the development of hypertension.
