目的通过观察白藜芦醇(Resveratrol,RES)对秀丽线虫衰老的影响,寻求延缓衰老的药物。方法秀丽线虫分为RES 0mg/L(对照组)、20mg/L、40mg/L、60mg/L、80mg/L组,观察秀丽线虫的寿命,以确定RES作用的最佳剂量;设立对照组(RES 0mg/L)和RES组(RES 60mg/L),评价RES对秀丽线虫寿命和生殖能力的影响;进行急性热应激实验、急性氧化应激实验,探讨RES延缓线虫衰老的机制。结果 60mg/L的RES是延长秀丽线虫寿命的最佳剂量;RES组的平均寿命长于对照组(P〈0.05);RES组生殖峰期产卵数量多于对照组(P〈0.01);RES组急性热应激实验中线虫平均生存时间和急性氧化应激实验中线虫平均生存时间均长于对照组(皆P〈0.01)。结论 RES能延缓秀丽线虫衰老,其机制可能与RES提高线虫的压力应激能力有关。
Objective To evaluate the anti-aging effect of Resveratrol(RES)on C. elegans in order to find anti- aging herbs. Methods Dosage-effect test:synchronized L4s were treated with RES 0 mg/L(control group) ,RES 20 mg/L,40 mg/L, 60 mg/L,80 mg/L groups,tested by lifespan assay to determine the effective dosages of RES. Life span analysis assay:synchronized L4s were treated as normal control group and RES group(RES 60 mg/L),tested by life span assay to evaluate the life-extension effect of RES. Fecundity test: RES treated and control animals' fecundity were tested to study effect of RES on the worms' reproduction. Stress resistance test: RES treated and control animals were treated by acute heat and oxidative stress to confirm the life span assay and explore the initial mechanism. Results 60 mg/L of RES was the best dosage to made C. elegans get the maximum lifespan. The mean lifespan of RES group was longer than the control group(P〈0. 05). Compared with the control group, peak fecundity of the RES group was significantly enforced (P〈 0, 01). RES lifted the stress resistance ability of C. elegans more significantly than the control group(P〈0.01). Coneluslons Resveratrol has anti-aging effect and the mechanism involved in elevation of the stress resistance ability of C. elegans.
