采用PCR-SSCP技术和DNA测序技术对124只贵州长顺绿壳蛋鸡的胰岛素生长因子1基因(IGF1)的In3片段进行多态性分析,并与体尺性状进行关联性研究,为贵州绿壳蛋鸡的遗传评估提供资料,并为选育提供有育种价值的分子标记。结果表明:试验鸡群的IGF1基因In3片段存在3个单核苷酸突变位点,分别为A(-981)G、A(-953)T和T(-881)C;该群体偏离Hardy-Weinberg平衡状态,且该位点处于高度多态(PIC〉0.5);F2F2基因型个体体斜长和胸深均低于其他基因型个体,推测该基因型可能是长顺绿壳蛋鸡向小型蛋鸡方向选育的有效分子标记。
In order to provide genetic evolution data and molecular markers for Guizhou blue-shell chickens breed-ing, PCR-SSCP and DNA sequencing technology were used to analyze the polymorphism and association of insulin-like growth factor (IGF1) gene on In3 fragment with body traits from 124 Changshun blue-shell chickens. The results showed that there were three nucleotide mutations, A ( -981 ) G, A (-953) T and T ( -141 ) C, in In3 fragment of IGF1 gene and the test groups were in high polymorphie state ( PIC〉0. 5 ). The distributions of genotypes were deviated from Hardy-Weinberg equilibrium ( P〈0.05 ). Association analysis indicated that the body slanting length and chest depth of the chick-ens with F2 F2 geuotype were both lower than those with other geuotypes, and the somatotype with F2 F2 genotype was rela-tively small, suggesting that genotype F2 F2 might be the marker for the breeding of Changshuu blue-shell chickens towards miniature laying chickens.
