本研究利用高效排阻色谱与多角度激光散射分析仪、示差折光仪串联建立淀粉结构分析的方法并得出相关分子特征参数。结果表明:采用Berry法三阶拟合可推导得出淀粉绝对分子质量及其分布、平均旋转半径(rz)、多分散系数。普通玉米淀粉、蜡质玉米淀粉、小麦淀粉、糯米淀粉、大米淀粉及马铃薯淀粉的重均分子质量Mw(rz)分别为17.94×10^7g/mol(79nm)、18.11×10^7g/mol(81.1nm)、7.86×10^7g/mol(50.4nm)、13.04×10^7g/mol(62.9nm)、64.3×10^7g/mol(64.3nm)及7.73×10^7g/mol(75.9nm)。该方法可作为一种简便、快速的测定淀粉分子结构的现代分析技术。
A method using high-performance size-exclusion chromatography (HPSEC) equipped with multi-angle laser-light scattering (MALLS) and refractive index detector (RID) was presented for determining molecular features of starch. Results indicated that the weight-average molecular weight (Mw), z-average radius of gyration (rz) and polydispersity index could be obtained using the Berry extrapolation method with a third-order polynomial fitting. The Mw (rz) of common maize starch, waxy maize starch, wheat starch, sticky rice starch, rice starch and potato starch were measured to be 17.94 ×10^7g/mol (79 nm), 18.11 ×10^7g/mol (81.1 nm), 7.86 ×10^7g/mol (50.4 nm), 13.04 ×10^7g/mol(62.9 nm), 64.3 ×10^7g/mol(64.3 nm), and 7.73 ×10^7g/mol (75.9 nm), respectively. This method can provide a convenient and easy approach for starch molecule structure analysis.
