该研究采用RashidNp-nPP比色法,通过单因素实验和正交实验对可生物降解聚丁二酸丁二醇酯(PBs)的铜绿假单胞菌产脂肪酶条件进行了优化。研究结果表明:最适培养条件为:温度29℃,摇床转速120r/min;最佳培养基为:蔗糖0.5%,硫酸铵0.05%,硫酸‘亚铁0.005%,Tween一60与Span-80按l:l复配乳化剂0.5%,接种量5%,培养基初始pH9.0。优化后酶活可达33.74184U/mL。
Rashid N p-nPP colorimetric method was used to demine the lipase activity, which was produced by Pseudomonas aeruginosa cultured under optimized conditions through mono-factor and orthogonal experiments. Results show the optimized conditions of culture as 29 ℃ and rotating speed 120 m; while the optimal culture media is sucrose 0.5%, (NI-L)2SO4 0.05%, FeSO4 0.005%, compound emulsifier (Tween--60 : Span-80 as i : 1 )0.5%, seed volume 5%, and the initial pH 9.0. After optimization,the lipase activity is up to 33.741 84 U/mL.
