目的 观察骨桥蛋白(Osteopontin,OPN)在寻常型银屑病皮损组织中的表达变化,并探讨其意义.方法 采用PV-9000免疫组织化学法检测20例寻常型银屑病皮损组织及15例健康对照皮肤组织中OPN的表达,在显微镜下选取阳性程度最高的“热点区域”,使用Image-Pro Plue软件计算两者灰度值并进行比较.结果 银屑病皮损中OPN为角质形成细胞的细胞质着色,为棕黄色,主要表达于表皮的基底层和棘层;健康对照皮肤组织中为黄色及淡黄色着色,主要位于基底层和棘层的下部.两者的灰度值分别为107.23 ±21.67和56.82±9.92(P <0.05).结论 OPN在寻常型银屑病皮损组织中表达上调,可能参与了银屑病的发病过程.
Objective To investigate the expression changes of osteopontin (OPN) in psoriatic lesions of psoriasis vulgaris and its significance. Methods PV-9000 immunohistochemical method was applied to detect the expression of OPN in both psoriatic lesions from 20 patients with psoriasis vulgaris and healthy controls from 15 healthy people. Hot spots of which showed the highest positive degree were selected under microscope to calculate the grey level by Image-Pro Plus software. The comparison of grey levels between psoriatic lesions and healthy controls was carried out by chi-square test. Results In psoriatic lesions, OPN was indicated by brown granules in cytoplasm, mainly expressed in basal layer and spinous layer of epidermis. Comparatively, in healthy controls, light yellow to yellow granules mostly presented in cyto- plasm of cells in basal layer and inferior parts of spinous layer. The grey level of psoriatic lesions was 107.23 ±21.67 while it was 56.82 ± 9.92 in healthy controls between which the difference was statistical significant ( P 〈 0.05 ). Conclusion The expression of osteopontin is up-regnlated in psoriatic lesions, which might be involved in the pathogenesis of psoriasis vulgaris.
