目的 研究60Coγ射线照射胶质瘤细胞U251对β3GnT8及多聚乳糖胺糖链的影响,为治疗恶性肿瘤和研究高效、低毒的辐射防护剂奠定实验基础.方法 采用0Gy、1Gy、5Gy和8Gy剂量的60Coγ射线辐照胶质瘤细胞U251,Western blotting检测60Coγ射线对β3GnT8蛋白水平表达的影响;流式细胞术分析60Coγ射线对多聚乳糖胺糖链表达水平的影响.结果 随着60Coγ射线辐照剂量的增加,β3GnT8蛋白和细胞表面多聚乳糖胺糖链表达水平逐渐降低.结论 60Coγ射线照射胶质瘤细胞U251对β3GnT8和多聚乳糖胺糖链表达均有影响,为研究高效、低毒的辐射防护剂奠定一些实验基础.
Objective To provide some experimental evidence for curing malignant turnout and researching highly active and low mammalian toxicity radioprotectant,and to study the effect of ^60Co γ-ray on the expression of β 3GnT8 and polylactosamine in human glioma cell line U251.Methods Using 0Gy, 1Gy,5Gy and 8Gy dose of ^60Co γ-ray to irradiate human glioma cell line U251,using western blotting to measure the β 3GnT8 protein expression level,and using flow cytometry to detect the effect of ^60Co γ-ray on the expression of polylactosamine. Results The expression of β 3GnT8 and polylactosamine decreased gradually as the dose of ^60Co γ-ray increased. Conclusion ^60Co γ-ray affected the expression of β 3GnT8 and polylactosamine in human glioma cell line U251. Providing some experimental evidence for researching highly active and low mammalian toxicity radioprotectant.
