目的:探讨加味二至方对便秘型肠易激综合征大鼠5-HT的影响,为临床应用加味二至方提供理论依据。方法:24只Wistar大鼠随机分为两组,第一组(A组)18只;第二组(B组)6只为正常组。A组大鼠用冰水灌胃法造模,造模成功后随机分为模型组、对照组、中药治疗组,对照组给予西沙比利灌胃;中药治疗组给予加味二至方煎剂灌胃。模型组及正常组正常饲食、饮水。第29天禁食18—24h,正常饮水。断头处死测指标。取A组和B组大鼠回盲部及结肠的石蜡切片,采用SABC免疫组织化学染色法。每例切片随机选择3个视野,光学显微镜下400倍观察,应用Image—ProPlus专业图像分析系统,选取下列测量参数进行半定量分析:两组大鼠回盲部及结肠5-HT光密度、积分光密度、面积的表达。结果:灌服中药组大鼠的回盲部和结肠组织的大鼠肠道5-HT光密度、积分光密度、面积的表达明显降低(P〈0.05)。结论:加味二至方能降低便秘型肠易激综合征大鼠肠道5-HT分泌,从而改善便秘症状。
Objective: To explore the effect of Modified Erzhi Prescription on 5-hydroxytryptamine in rats with irritable bowel syndrome of constipation, to provide a theoretical basis for clinical application of Modified Erzhi Prescription, Methods: 24 Wistar rats were randomly divided into two groups, 18 for the first group ( Group A) ; 6 for the second group ( Group B) : normal group. The rats were fed with cold water to make models, after the success of modeling, they were randomly divided into the model group, control group, Chinese medicine treatment group. The control group were given gavage of eisapride; Chinese medicine treatment group were given gavage of Modified Erzhi Prescription decoction. The model group and normal eontrol group were fed normal food and water. They were fasted 18 -24 h only with normal water drinking on 29th day. The indicators were measured after being decapitated. The ileoceeal and colonic paraffin slices of group A and group B were taken to measure with SABC immunohistochemical staining. Three cases of vision for each slice were randomly selected to be observed under 400 times of optical microscope. The following parameters were selected to be measured by semi-quantitative analysis with professional Image-ProPlus image analysis system: rat ileoeecal and colonic 5 - HT optical density, integral optical density and area expression of two groups. Results : The rat intestinal 5 - HT optical density, integrated optical density, area expression of ileocecal and colonie tissues of Chinese medicine treatment group were significantly lower than those of other groups ( P 〈 0.05 ). Conclusion: Modified Erzhi Preseription can reduce the intestinal 5 - HT secretion of rats with irritable bowel syndrome of constipation, thereby impro- ving the symptoms of constipation.
