目的:采用制备单柱配合前后处理工艺来分离得到高纯度异甘草素。方法:采用正交设计优化前处理超声提取的条件,提取物经2次制备单柱梯度洗脱分离,浓缩后得目标产品。结果:超声提取最佳条件为:配制原料的质量浓度为30g/L,采用乙醇做提取溶剂,超声提取时间为60min,提取温度为20℃。选择适合的单柱进样浓度为100g/L,进样体积为60mL。单柱产品经浓缩得到质量分数在99%以上的异甘草素。结论:以柱色谱分离为核心,结合原料前处理和产品后处理工艺分离提纯异甘草素工艺简单易行,所得产品质量分数高。
Objective:High purity of isoliquiritigenin was obtained by preparative column combined with the pretreatment and post treatment.Methods:Orthogonal design was used to optimize the pre-treatment ultrasonic extraction conditions.The extract was separated two times by single column with gradient elution,then concentration was performed and the product was obtained.Results:The optimum conditions for extraction are as follows:the concentration of raw materials was 30g/L with ethanol as extraction solvent,extraction time was 60 min,and extraction temperature was 20℃.The suitable injection concentration and injection volume were 100g/L and 60mL,respectively.Finally,more than 99% purity of isoliquiritigenin product was obtained.Conclusion:The separation and purification of isoliquiritigenin is feasible by the core of column chromatography combined with pretreatment of raw material and post treatment of the product,the process is simple,and the purity of the product was high.
