目的:探讨干扰素-α联合地塞米松对神经细胞p11表达的影响.方法:神经细胞经过10-7mol/L地塞米松或和1 000 U/ml IFN-α处理后,利用RT-PCR检测各组p11 mRNA的表达水平;Western blot法检测p11蛋白的表达水平;免疫荧光检测各组p11蛋白的表达水平.结果:Real-time PCR 和Western blot结果提示1 000 U/ml IFN-α刺激组p11的表达降低,10-7 mol/L地塞米松刺激组p11的表达升高;IFN-α和地塞米松联合刺激组与对照组相比无显著差异.免疫荧光结果也显示IFN-α刺激组可抑制p11蛋白的表达,p11在IFN-α联合地塞米松组中的表达低于地塞米松组.结论:IFN-α可以下调神经细胞p11表达;也可以阻断地塞米松对p11表达的上调作用.
Objective:To investigate the effects of interferon-α combined with dexamethasone on pl 1 expression in nerve cell line. Methods: After treatment with 10-7 mol/L dexamethasone (DEX) or/and 1 000 U/ml interferon-α in nerve ceils, the expres- sions of pl 1 mRNA and protein in each group were determined using RT-PCR and Western blot respectively. Immunofluorescence was also performed to detect pl 1 expression. Results: RT-PCR and Western blot results showed that pl I expression was decreased in 1 000 U/ml interferon-α treatment group and increased in DEX treatment group. There was no significant difference in pl 1 expression be- tween combination group and control group. Immunofluorescence showed that pl 1 expression was inhibited in interferon-α treatment group and its expression in the combination group was decreased compared with DEX treatment group. Conclusion: Interferon-α could inhibit the expression of pl 1 and block dexamethasone-induced increase of pl 1 expression in nerve cells.
