从活性污泥中筛选出高效脱氮除磷菌,并进行分类鉴定,同时研究了不同pH和接种量对生长的影响.通过聚磷菌的富集培养、缺磷和富磷培养的筛选、PHB和异染颗粒染色实验进行鉴定,并通过除磷除氮实验、形态学和生理生化实验进行初步研究,通过16S rRNA进行分子鉴定.筛选得到一株高效脱氮除磷菌.该菌株在pH5-10之间均能正常生长,最佳生长pH为7.接种量在10%时生长效果最好,其中对氮磷的去除率分别达78.3%、87.8%.该菌株可以同时对氮和磷有较好的去除效果,通过形态学观察、生理生化试验和16S rRNA基因序列分析,构建系统发育树,初步鉴定为肠杆菌属.
The highly effective denitrification and phosphorus removal bacteria was screened from activated sludge, it was classified and identified too. At the same time, the effects of pH and inoculation amount on the growth were studied. Getting one strains by phosphorus enrichment culture, and PHB and 16S rRNA staining, and carrying out the study of nitrogen and phosphorus removal rate, the morphological and physiological and bi- ochemical experiments. A highly efficient nitrogen and phosphorus removal bacteria was screened. The strain was able to grow in pH 5-10, pH7 was the best. The best inoculation amount to the growth was 10%. The rates of nitrogen and phosphorus reached 78.3 %, 87.8 %. The strain can remove nitrogen and phosphorus at the same time, and it can identified as Lactobacillus genus by morphological observation, physiological and biochemical test, and 16S rRNA gene sequence analysis and construction of phylogenetic tree.
