多酚是桑椹中的重要抗氧化活性成分之一,建立高效简便提取桑椹多酚的工艺技术,有助于研制与开发具有药用保健功效的桑椹多酚产品。以新鲜成熟桑椹为原料,采用超声波辅助乙醇提取法提取桑椹多酚。在单因素试验的基础上,以桑椹多酚提取得率为考察指标,采用Box-Benhnken中心组合试验设计法优化提取工艺条件,建立了包括乙醇体积分数、提取溶液pH值、原料质量浓度、超声波功率和提取时间5个因素的桑椹多酚提取得率回归模型。利用Design-Expert软件对数据进行回归分析并结合验证试验结果,确定最佳提取工艺条件为:乙醇体积分数60.0%,提取溶液pH 5.0,原料质量浓度0.025g/mL,超声波功率405 W,提取时间30 min。在此工艺条件下提取桑椹多酚的实际得率为3.016%,预测值为3.161%。建立的回归模型可以较好地预测桑椹多酚提取得率,优化的提取工艺条件具有一定的实用价值。
Polyphenols are one of the important components in mulberry fruit with antioxidant activity. To establish efficient and easy-operated extracting technology for mulberry polyphenols will facilitate development of mulberry polyphenol products for medical and healthcare uses. Fresh mature mulberry fruit was used as experimental material to extract polyphenols by ultrasonic-assisted ethanol extraction method. Based on single-factor experiments,the effect of ethanol concentration,extraction pH value,mass concentration of raw material,ultrasonic power,and extraction time on the yield of polyphenols from mulberry fruit were optimized by response surface methodology. Combined with regression analysis by Design-Expert software and validation tests,the optimized extraction process was determined as follows: ethanol concentration 60. 0%,extraction pH value 5. 0,mass concentration of raw material 0. 025 g /mL,ultrasonic power 405 W,and extraction time 30 min.Under this process condition,the yield of polyphenols from mulberry fruit was 3. 016%,and the predictive value was 3. 161%. The result indicated that the establishedmodel could be used to predict the extraction yield of polyphenols from mulberry fruit,and the optimized extraction process condition has certain practical value.
