目的利用阿尔茨海默症(Alzheimer’s disease,AD)模型构建Slit2蛋白过表达杂合子小鼠,并进行鉴定,为进一步在动物整体水平研究Slit2蛋白过表达在阿尔兹海默症中的作用提供研究平台。方法将AD模型鼠Tg2576的杂合子小鼠(Tg2576^+/-小鼠)进行繁殖并鉴定,然后将鉴定结果为阳性的子代Tg2576^+/-小鼠与Slit2蛋白过表达的Slit2-Tg纯合子小鼠进行杂交,鉴定其子代小鼠的基因型。结果成功繁育Tg2576^+/-小鼠和Slit2-Tg小鼠,并通过鉴定得到Tg2576^+/-小鼠,与Slit2-Tg小鼠杂交并繁殖,获得基因型为Tg2576^+/-;Slit2-Tg小鼠5只。结论本研究利用AD模型鼠成功构建了Slit2蛋白过表达杂合子小鼠,为进一步研究Slit2蛋白过表达在AD发生发展中的作用提供了良好的动物模型。
Objective To generate and identify the heterozygous mouse model of Alzheimer's disease (AD) by hybridization with mice over-expressing Slit2 protein, and to serve the research of the role of over-expression of Slit2 protein in AD at the whole-animal level. Methods The heterozygous Tg2576 transgenic mice were generated and identified. The positive offsprings were mated with Slit2-Tg homozygous mouse, in which Slit2 protein was over-expressed. The gene type of the offsprings was identified. Results The Tg2576 heterozygous mice and Slit2-Tg mice were successfully generated and Tg2576 ^+/- mice were identified. Five Tg2576 ^+/-; Slit2-Tg mice were obtained through mating of Tg2576 ^+/- mice with Slit2-Tg mice. Conclusions Slit2 protein over-expressing heterozygous mice models are successfully generated by hybridization with AD model mice, which can supply a novel animal model for further research of the Slit2 protein over-expression in Alzheimer's disease.
