目的观察三七总皂苷对糖尿病大鼠早期视网膜电图(ERG)及视网膜组织胶质纤维酸性蛋白(GFAP)表达的影响。方法采用链脲佐菌素(65mg·kg^-1)一次性腹腔注射SD大鼠建立糖尿病大鼠模型,随机分为模型组、三七总皂苷组(35mg·kg^-1)及导升明组(167mg·kg^-1),另设正常对照组。灌胃给药,20周后测定ERG后应用免疫组织化学法、蛋白印迹和逆转录PCR法检测大鼠视网膜中GFAP蛋白及mRNA的表达。结果糖尿病大鼠视网膜ERGa波、b波和OPs振幅降低,a波潜伏期延长,与正常组比较差异有统计学意义(P〈0.05)。三七总皂苷组大鼠视网膜ERGb波和OPs的振幅明显升高,与模型组比较差异有统计学意义(P〈0.05)。同时,与正常组相比,糖尿病大鼠视网膜GFAP蛋白及mRNA的表达明显增加;与糖尿病模型组相比,三七总皂苷可降低糖尿病大鼠视网膜中GFAP蛋白及mRNA的表达(P〈0.05)。结论三七总皂苷能够保护糖尿病大鼠视网膜,这一作用可能是通过降低视网膜GFAP的表达来实现的。
Objective To observe the effect of Panax Notoginseng Saponins (PNS) on electroretinogram (ERG) and glial fi- brillary acid protein (GFAP) in retina of diabetic rats, and to explore the mechanism of PNS on diabetic retinopathy. Methods A single intraperitoneal injection of streptozotocin (65 mg · kg^-1)was used to induce diabetic rats. Model rats were divided into diabetes mellitus (DM) group, PNS group and Doxium group. The normal group and model group were given normal water, and PNS group and Doxium group were given PNS (35 g · kg^-1) and Doxium (167 mg · kg^-1) through intragastric administration. Electroretinogram examinations were performed after the experiment, and the retinal sections were observed the expression of GFAP by immunological histochemistry as- say and western blot. The expressive abundance of GFAPmRNA were detected by RT - PCR. Results Compared with control group, there was significantly decreased in the amplitudes of a wave, b wave and oscillatory potentials (OPs) after experiment in diabetic group (P 〈 0.05 ). The PNS group were improved markedly (P 〈 0. 05 ). The expression of GFAP protein and GFAPmRNA in the retina were significantly higher in DM rats than were in control group ( P 〈 0.05 ). Compared with the model group, the expression of GFAP in PNS group decreased evidently ( P 〈 0.05 ) and the expression of GFAP mRNA were lower than those in model group ( P 〈 0.05 ). Conclusion PNS can protect the early retinopathy in diabetic rat and inhibit the expression of GFAP in retina tissue.
