目的探讨转染腺病毒载体对大鼠催乳素细胞增殖功能的影响及作用机制。方法应用BrdU标记法检测转染腺病毒载体后大鼠垂体催乳素细胞的增殖功能,免疫荧光方法和荧光素酶报告基因分别检测转染腺病毒载体的催乳素细胞中磷酸化cAMP反应元件结合蛋白(pCREB)和cAMP反应元件(CRE)的表达。Westernblot方法检测腺病毒载体对垂体前叶细胞中增殖细胞核抗原(PCNA)表达的影响。结果转染腺病毒载体的催乳素细胞组较未转染组增殖功能明显增强(P〈0.01),pCREB蛋白和CRE元件的表达均明显增加(P〈0.01),PCNA蛋白的表达亦明显增强(P〈0.01)。结论应用腺病毒载体转移基因将增加大鼠催乳素细胞的增殖功能,其细胞内信号转导机制可能与腺病毒载体诱导pCREB、PCNA蛋白的表达增强有关。
Objective To investigate the effects of adenovirus vectors on the proliferation of rat pituitary lactotrophs.Methods Influence of adenovirus vectors on the proliferation of pituitary lactotrophs was detected by BrdU labelling.Expression of pCREB protein and CRE in the lactotrophs transfected by adenovirus vectors was determined by immunofluorescence method and luciferase reporter assay.Western Blot method was employed to detect the expression of PCNA protein in the pituitary cells transfected by adenovirus vectors.Results The basal proliferation of lactotrophs in the adenovirus groups was enhanced significantly compared with the control group(P 〈 0.01).After transfected with adenovirus vector,the expression of pCREB,PCNA protein and CRE gene was significantly up-regulated in the lactotrophs (P 〈 0.01).Conclusion Adenovirus transgene expression increases the proliferation of the rat lactotrophs,the up-regulation of pCREB and PCNA may be involved in this process.
