目的:研究白血病抑制因子(leukemia inhibitory factor,LIF)信号通路的相关分子在人牙周组织的表达。方法:在体内研究部分,取肿瘤患者扩大切除的带颌骨健康牙,免疫组化法研究LIF信号通路相关分子(LIF、LI-FR、Jak2、p-Jak2、Stat3和p-Stat3)在牙周组织的表达和定位。在体外研究部分,取正畸牙,体外培养牙周膜细胞,利用人重组LIF刺激牙周膜细胞,免疫荧光法观察p-Jak2和p-Stat3的表达情况。结果:人牙周组织LIF、LIFR、Jak2、p-Jak2、Stat3和p-Stat3均呈阳性,其中牙周膜细胞、成骨细胞和成牙骨质细胞呈LIF、LIFR、Jak2和p-Jak2胞浆着色,Stat3和p-Stat3为胞核着色。在人重组LIF作用下Jak2和Stat3发生磷酸化,p-Stat3部分入核。结论:LIF信号通路的相关分子在人牙周组织呈阳性表达,LIF可激活牙周韧带细胞中Jak2-Stat3信号轴。
Objective:To evaluate the expression of moleculars of LIF signaling in the human periodontal tissue.Methods:For the in vivo study,the madibular bone with the teeth was cut from the patient who was subjected to the madibular tumor expanded excision surgery.The expressions of LIF,LIFR,Jak2,p-Jak2,Stat 3,and p-Stat3 in the periodontal tissue were investigated with the immunohistochemistric analysis.For the in vitro study,the periodontal ligament cells were harvested from the extracted teeth for orthodontic treatment.The cells were treated with or without human recombinant LIF.The expressions and nuclear translocation of p-Jak2 and p-Stat3were tested with immunofluerescence.Results:All the LIF signaling moleculars were positively stained in human periodontal tissue.LIF,LIFR,Jak2,and p-Jak2 were expressed in cytoplasm of the periodontal ligament cells,osteoblasts,and cementoblasts,while Stat3 and p-Stat3 in cytoplasm and nuclears of these cells.When treated with LIF protein,the phosphorated Jak2 and Stat3 were unregulated and p-Stat3 was translocated to nuclear.Conclusion:All the LIF signaling moleculars were positively stained in human periodontal tissue.LIF can activated the Jak2-Stat3 axis in the periodontal ligament cells.
