[目的]探讨壳聚糖-乳糖酸-超顺磁性氧化铁纳米颗粒(CS-LA@SPION)应用于磁共振分子成像的可行性.[方法]制备壳聚糖(CS)与乳糖酸(LA)修饰的超顺磁性氧化铁纳米颗粒(SPION),采用透射电子显微镜与动态光散射纳米粒度分析仪进行形貌表征;MTT法检测细胞毒性;裸鼠尾静脉注射给予CS-LA@SPION后利用体内磁共振成像(MRI)验证磁性纳米粒对肝脏的靶向增强作用.[结果]给予CS-LA@SPION后30min,1,2h时肝脏T2值分别下降68.6%,78.9%,61.8%,注射后1h肝脏T2WI信号下降最显著.[结论]CS-LA@SPION对肝脏具有特异性靶向增强作用,可作为肝脏MRI靶向对比剂.
OBJECTIVE To investigate the feasibility of the chitosan-lactose acid-superparamagnetic iron oxide nanoparticles(SPION)used in magnetic resonance molecular imaging.METHODS Preparation of chitosan and lactose acid modified SPION(CS-LA@SPION)was characterized by transmission electron microscopy and dynamic light scattering nanometer particle size analyzer,and cell cytotoxicity was observed by MTT assay,and after injecting the CS-LA@SPION to nude mice tail vein,the nanoparticles for liver targeting enhancement were verified by MRI in vivo.RESULTSafter injecting with CS-LA@SPION at 30 min,1h,and 2h,liver T2 WI signal decreased 68.6%,78.9% and 61.8%,respectively.CONCLUSION CS-LA@SPION has specific target effect to liver cell and could be used as liver-targeted MR contrast agent.
