目的:研究精子顶体相关基因DKKLl在正常人和男性不育患者睾丸组织中的表达差异。方法:采用RT—PCR和WesternBlot方法检测DKKLl基因在人多组织中的表达特征,并比较其在正常人和男性不育患者睾丸组织中的表达差异。采用免疫组化方法分析DKKLl蛋白在正常和男性不育患者睾丸组织中的表达差异。结果:人多组织RT—PCR和WesternBlot结果显示DKKLl基因为睾丸组织特异性表达,与正常人睾丸组织比较,DKKLl基因在男性不育患者睾丸组织中表达减弱或消失。免疫组化结果显示DKKLl蛋白主要定位在正常男性睾丸的精母细胞和圆形精子细胞,而在唯支持细胞综合征(SCOS)和隐睾患者睾丸组织中不表达,在各种生精细胞阻滞患者的睾丸中观察到不同的表达特征。结论:DKKLl基因在男性不育患者睾丸组织中对精子发生可能具有重要作用.其表达降低或消失可能是男性不育症发生的重要原因之一。
Objective To investigate the expression difference of Dickkopf-like 1 Gene (DKKL1) in testis tissue of normal and male infertile patients. Methods The characristics of DKKL1 gene expression in multi-tissues in human was detected and compared by RT-PCR and Western Blot. The localization of DKKL1 protein in normal and male infertility testis was assesed by immunohistochemistry. Results In human multi-tissues, DKKL1 gene expressed exclusively in testis. Compared with the normal human testis, DKKL1 gene expression of male infertility decreased or disappeared. Additionally, the DKKL1 protein was predominantly located in spermatocytes and round spermatids in normal testis while it did not expressed in the patients with sertoli cell only syndrome and the infertile male patients. The different characristics of DKKL1 gene expression were observed in patients with spermatogenic arrest. Conclusion DKKL1 gene may play an important role in male infertility mammalian spermatogenesis, and its abnormal expression may be one of the essential factors in male infertility.
