目的 以损伤转基因斑马鱼胚胎为模型研究淫羊藿苷对受损血管发生的影响.方法 采用血管内皮生长因子受体抑制剂(VRI)制备斑马鱼胚胎的血管损伤模型,3h后给予不同浓度(30 100,300 μM)的淫羊藿苷处理24、48 h,之后对胚胎体节间血管和肠下静脉进行拍照并计数.结果 给予VRI作用3h后观察到受损血管数量不到10条.3个浓度的淫羊藿苷均可促进受损的斑马鱼血管发生,其促血管发生的作用在48 h时较为明显,100、300 μM的淫羊藿苷组体节间血管数量分别为22条和26条左右.结论 淫羊藿苷可促进受损的斑马鱼体节间血管和肠下静脉的发生.
Objective To investigate the effect of icariin on impaired vessel using impaired transgenic zebra fish embryo model. Methods Zebra fish embryo vessels impaired model was induced by vascular endothelial growth factor receptor inhibitor. Three hours later, these zebra fish embryos were treated with different concentrations of icariin (30, 100, 300μM) for 24 hours or 48 hours, then intersegmental blood vessels and subintestinal veins of embryos were counted and photographed respectively. Results There were less than 10 defective vessels in the group treatment with VRI for 3 h. All three concentrations of icariin could improve the growth of impaired vessels of zebra fish embryos, and the effect was more significance on 48 hours. The number of intersegmental blood vessels in 100 and 300 μM icariin groups were about 22 and 26, respectively. Conclusion Icariin has the angiogenesis - facilitated effect on intersegmental blood vessels and subintestinal veins in the impaired - vessel of zebra fish embryo.
