目的:观察丹栀逍遥散对多囊卵巢综合症(PCOS)大鼠差异蛋白质表达谱的影响。方法:应用皮下埋植左旋18-甲基炔诺酮硅胶棒联合人绒毛膜促性腺激素注射复制PCOS模型;双向电泳技术分离差异蛋白表达谱,基质辅助激光解吸/电离飞行时间质谱(MALDI-TOF MS)鉴定差异蛋白点;免疫组化、蛋白免疫印迹和荧光定量PCR验证结果。结果:丹栀逍遥散3.75g/kg显著改善大鼠卵巢增大及卵泡扩张,质谱成功鉴定8个蛋白点,其中点5为β抑制蛋白2(β-arrestin 2,β-arr2),β-arr2在模型组显著降低,丹栀逍遥散显著增加其表达(P〈0.01)。结论:丹栀逍遥散通过增加β-arr2表达,调控β-arr2介导的信号通路防治PCOS。
Objective: To identify the underlying mechanisms in the protective effects of Danzhixiaoyao Powder(DZXYP) on polycystic ovary syndrome rat model.Methods: PCOS was introduced by subcutaneous implantation of 18-methyl norethisterone silica gel stick combined with subcutaneous injection of human chorionic gonadotrophin.Two-dimensional polyacrylamide gel electrophoresis(2-DE) and matrix-assisted laser desorption/ionization time-of-flight mass spectrometry(MALDI-TOF MS) were used to identify differentially expressed proteins.Immunohistochemistry,real time quantitative polymerase chain reaction(RQ-PCR) and western blot were performed to verify the results above.Results: DZXYP ameliorated ovary enlargement and follicle dilatation.Among eitht successfully identified proteins that were differentially expressed,β-arrestin 2(β-arr2) was upregulated in DZXYP pretreated PCOS group when compared with PCOS group.Conclusion: DXR could alleviate PCOS which might be related to increased expression of β-arr2.
