目的考察在全氟异丁烯(PFIB)吸入毒理实验中引进清醒动物肺功能检测系统是否有助于PFIB中毒的诊断和治疗。方法利用清醒动物肺功能检测系统,检测PFIB吸人中毒大鼠中毒前后不同时间的呼吸功能指标(吸气时间、呼气时间、呼吸频率、吸气流量峰值、呼气流量峰值、潮气量和呼气量)。结果Wistar大鼠给予PFIB150mg/m3暴露吸入8min。4h后即观察到吸气时间、呼气时间明显缩短,8h后两者分别降至129ms和215ms,并持续至24h;在染毒后8h吸气流量峰值和呼气流量峰值显著升高,呼吸频率明显加快,潮气量和呼气量明显降低。结论清醒大鼠肺功能检测系统操作简便快捷,数据稳定。灵敏度高,可及时有效检测出PFIB吸人中毒大鼠呼吸功能指标的变化,可广泛应用于吸入毒理药理实验研究中。
Objective To measure if the pulmonary function test system of unrestrained animals could help diagnosis and treatment of perfluoroisobutylene (PFIB) inhalation toxieosi Methods To record the change in pulmonary function parameters, such as inspiratory time (TI), expiratory time (TE), breathing frequency (F), peak inspiratory flow (PIF), peak expiratory flow(PEF), tidal volume (VT) and expired volume (VE), by unrestrained whole body plethysmography (EMKA Technologies, France), before and after Wistar rats exposed to 150 mg/m3 PFIB for 8 min. Results The TI and TE decreased obviously at 4 h after PFIB exposure, and reduced to 129 ms and 215 ms at 8 h after PFIB exposure, respectively. The PIF, PEF and the breathing frequency increased significantly, on the contrary, the VT and VE decreased markedly at 8 h after PFIB ex- posure. Conclusion The pulmonary function test system can be handled easily, and can be used for measuring the changes in pulmonary parameters in the study of inhalation toxicology.
