目的以胶原诱导性关节炎(CIA)大鼠为模型,观察沙利度胺对CIA鼠关节滑膜组织中单核细胞趋化蛋白(MCP-1)的影响。方法利用牛Ⅱ型胶原建立Wistar大鼠CIA模型,造模成功后随机分为正常对照组、生理盐水CIA模型对照组、吲哚美辛阳性药物对照组和沙利度胺治疗组。结果在胶原第1次免疫后第11~13天,CIA鼠模型建立,关节肿胀在第17天达高峰,沙利度胺给药后能显著降低足肿胀的程度,减轻关节炎症评分指数,抑制炎症细胞浸润,减少滑膜细胞增生,并减少鼠滑膜组织中MCP-1的表达。结论沙利度胺的抗炎作用可能与抑制关节滑膜组织中趋化因子MCP-1的表达相关。
Objective To observe the effect of thalidomide on MCP-1 in synovial joints of rats. Methods A CIA model of Wistar rats was established by injecting bovine Ⅱ collagen (BIIC). The rats were randomly divided into normal control group, CIA control group, indometacin group and thalidomide group. Results The CIA model was successfully established 11 - 13 days after injection of BIIC. The swelling of joints reached its peak 17 days after injection of thalidomide and was milder in thalidomide group. Thalidomide significantly ameliorated the symptoms of CIA rats, inhibited swelling of joints, and reduced inflammatory invasion and expression of MCP-1. Conclusion The anti-inflammatory action of thalidomide is related to the expression of MCP-1.
