探讨PKM2在白血病NB4细胞中的分布情况及对c-Myc蛋白的影响。免疫荧光技术检测PKM2在NB4细胞中的分布情况。将含靶点序列的GV248载体、包装质粒Helper1.0和Helper2.0共转染293T细胞,包装慢病毒。收集病毒上清,浓缩纯化后感染NB4细胞,荧光显微镜观察感染效率,Western blotting检测PKM2和c-Myc蛋白的表达变化。CCK-8实验检测细胞增殖情况。结果显示,PKM2在NB4细胞核及细胞质中均有表达,细胞核中比例较大。成功构建LV-PKM2-RNAi病毒,感染的细胞PKM2和c-Myc表达下调(p〈0.05),细胞增殖能力降低(p〈0.05)。这些结果表明PKM2主要分布在NB4细胞核,抑制PKM2表达可明显抑制NB4细胞增殖,并抑制c-Myc蛋白表达。
This study aimed to investigate the distribution of PKM2 in leukemia NBB4 cells and its effect on c-Myc protein. The distribution of PKM2 protein in NB4 cells was detected by immunofluorescence assay. The GV248 vector, packaging plasmid Helper 1.0 and Helper 2.0 were co-transfected into 293T cells to package lentivirus. The viral supernatant was collected and purified to infect NB4 cells, and the infection efficiency was observed by fluorescence microscopy. The expression of PKM2 and c-Myc was detected by Western blotting. CCK-8 assay was used to examine cell proliferation. The results showed that PKM2 protein was expressed in the nucleus and cytoplasm of NB4, and the prop ortion was larger in nucleus. Recombinant lentivirus LV-PKM2-RNAi was successfully constructed, and the expression of infected PKM2 and c-Myc cell was decreased (p 〈0.05), and the proliferation of NB4 cells infected with LV-PKM2-RNAi was inhibited. These results indicated that PKM2 was mainly distributed in the nuclear of NB4 cell, and the inhibition of PKM2 could inhibite the proliferation of NB4 cell and the expression of c-Myc.
