目的制备包裹吲哚菁绿(ICG)全氟己烷(PFH)的纳米级双模态显像造影剂,观察其体外热致相变,光声成像及超声成像效果。方法采用多步乳化技术制备包裹ICG及PFH的聚乳酸-羟基乙酸共聚物PLGA纳米粒(INP纳米粒),观察及分析INP纳米粒理化性质、相变过程及体外增强光声及超声显像效果。结果成功制备INP纳米粒,其平均粒径为(607.26±23.53)nm,温度达70℃时,纳米粒逐渐增大变为微气泡,温度保持不变随着时间延长,气泡变大且数量增多,最后气泡破裂,整个过程约持续40s。激光辐照30s后,INP纳米粒可检测到明显光声信号和超声信号。结论成功制备包裹ICG和PFH的PLGA纳米粒,其在激光辐照下可产生光声信号和超声信号。
Objective To prepare the dual-mode nanoparticle contrast agent loaded with indocyanine green and perfluorohexane(PFH),and to observe its phase-transition by heating,photocoustic and ultrasound imaging in vitro.MethodsThe PLGA nanoparticles loaded with ICG and PFH(INP nanoparticles)were prepared with multi-emulsion technique,and then the physicochemical properties,phase-transition on heatingplate and photoacoustic/ultrasound imaging were observed.Results The INP nanoparticles were prepared successfully with diameter of(607.26±23.53)nm.The phase-transition of the nanoparticles occurred when the temperature of heatingplate showed 70℃,and the size and the number of the bubbles increased with the time until eruption,which lasted for 40 s.After irradiation with laser for 30 s,the photoacoustic and ultrasonic signals of INP nanoparticles were obviously detected.Conclusion The PLGA nanoparticles loaded with ICG and perfluorohexane are successfully prepared,which can produce photoacoustic and ultrosonic signal under irradiation with laser in vitro.
