目的:研究不同脱色方法对全真一气汤多糖含量的影响,选择适宜的脱色方法,并初步探讨其对免疫抑制小鼠外周血免疫功能的影响。方法:分别采用大孔树脂静态吸附法、活性炭吸附法、过氧化氢氧化法进行脱色,以色素脱除率和多糖含量为指标,进行脱色效果比较;采用环磷酰胺造成免疫抑制模型,观察全真一气汤多糖对小鼠外周血白细胞、淋巴细胞及IL-2的影响。结果:大孔树脂静态吸附法处理后,多糖的色素脱除率为(48.32±1.05)%,多糖含量(39.63±1.08)%,相比粗多糖,含量有所提高。活性炭和过氧化氢处理后,色素脱除率虽然分别达到(68.49±1.02)%,(68.98±0.69)%,但多糖含量降低;全真一气汤多糖可对抗免疫抑外周血白细胞及淋巴细胞数目的降低,刺激外周血血清IL-2分泌的增加。结论:大孔树脂法适宜用于全真一气汤多糖脱色素;全真一气汤多糖可以提高免疫抑制小鼠的免疫功能。
Objective:To investigate the effect of different deeolorization methods on the polysaccharide content of Quanzhen Yiqi Decoction and select a suitable odeeolorization method. And discussed its effect on the peripheral immune function in immunosuppressive mice. Methods : Three routine decolorization methods were compared, state macroporousresin absorption, activated carbon absorption and xidation by hydroge eroxide,with the indicators of decolorization percentage and the content of polysaccharide. Using cyclophosphamide to buld immunosuppression model observe the effect of Quanzhen Yiqi Decoction on peripheral blood leukocytes, lymphocytesand the IL- 2 inimmunosuppressive mice. Results:For state macroporousresin absorption,the decolorization percentage and polysaccharide content were 48.32 ± 1.05% and 39.63 ± 1.08%. The decolorization percentages were 68.49 ±1.02% and 68.98 ±0.69% for activated carbon absorption and oxidation by hydroge eroxide but the polysaccharide contents were too low. Quanzhen Yiqi Decoction can fight against the reduce of the number of peripheral blood leukocyte and lymphocyte in immunosuppression mice, to stimulate the increase of peripheral blood serum IL- 2. Conclusion:The state macroporousresin absorption was suitable for the decolorization of the polysaccharide of Quanzhen Yiqi Decoction;Quanzhen Yiqi Decoction can improve the immune function of immunosuppressed mice.
