采用简并引物扩增得到瓦氏黄颡鱼肝型和心型脂肪酸结合蛋白eDNA片段,长度分别为335、425bp;获得的111、114个氨基酸残基与鲤鱼(Cyprinus carpio)肝型、斜齿鳊(Rutilus rutilus)心型脂肪酸结合蛋白的氨基酸序列同源性分别达到81%和85%,表明克隆所得序列均为瓦氏黄颡鱼脂肪酸结合蛋白.定量PCR分析表盟,肝脏和腹腔脂肪组织均是肝型和心型脂肪酸结合蛋白表达的主要场所.
The liver and heart fatty acid binding-protein (FABP) were amplified with degenerated primers from darkbarbel catfish,Pelteobagrus vachelli. Part of the liver FABP consisted of 335 bp encoded a protein of 111 amino acids, and had an overall identity of 81% in comparison with that of Cyprinus cario. Part of the heart FABP consisted of 425 bp with 26 bp 3'-untranslated region, encadeda protein of 114 amino acids, had an overall identity of 85% in comparison with that of Rutilus rutilus. Quantitative PCR analysis showed that liver and heart FABP were mainly expressed in liver and adipose tissues.
