目的用细胞膜绿色荧光探针Di O标记人骨髓间充质干细胞(h BMMSCs),探讨其对h BMMSCs生物学功能的影响。方法采用Ficoll密度梯度离心法分离、培养h BMMSCs;流式细胞术检测Di O对h BMMSCs的标记效率;通过Transwell迁移实验、平板克隆实验、生长曲线分析、成脂及成骨诱导实验分别检测Di O对h BMMSCs迁移、增殖和多向分化能力的影响。结果Di O对h BMMSC的标记效率可达(99.5±0.4)%;Transwell迁移实验、平板克隆实验、生长曲线分析、成脂及成骨诱导实验结果表明,与未标记h BMMSCs相比,标记后的h BMMSCs迁移、增殖及成脂、成骨分化能力未见明显改变,差异均无统计学意义(P均〉0.05)。结论经Di O处理未见明显影响h BMMSCs的生物学功能,可做为安全、有效的h BMMSCs体外标记绿色荧光染料。
Objective To investigate the possible effects of cell membrane green fluorescent probe 3,3'-dioctadecyloxacarbocyanine perchlorate( Di O) on the biological function of human bone marrow mesenchymal stem cells( h BMMSCs) after labeling h BMMSCs in vitro. Methods h BMMSCs were separated by Ficoll density gradient centrifugation,and then labeled by Di O. The labeling efficiency of Di O was detected by flow cytometry. The effects of Di O on the migration,proliferation and multi-directional differentiation potential of h BMMSCs were analyzed by Transwell migration assay,colony formation assay,growth curve analysis,and adipogenic and osteogenic induction assay,respectively. Results The efficiency of Di O labeling h BMMSCs reached to( 99. 5 ± 0. 4) %. There were no significant differences in the migration, proliferation, and adipogenic and osteogenic differentiation potential between Di O-labeled h BMMSCs and unlabeled h BMMSCs( P〉0. 05). Conclusion Di O labeling h BMMSCs does not affect their biological function,indicating that Di O may be a safe and effective green fluorescent dye for labeling h BMMSCs in vitro.
