目的研究种植内皮祖细胞对小径聚氨酯人工血管移植通畅率的影响,探讨改善小径人工血管通畅率的新策略。方法诱导健康成人外周血的单个核细胞分化成内皮祖细胞。种植内皮祖细胞到小径聚氨酯人工血管表面后,用酶联免疫吸附法测定其分泌组织纤溶酶原激活物(t-PA)和一氧化氮(NO)水平。13只家犬随机分为两组,将实验组6只种植内皮祖细胞的小径聚氨酯人工血管和对照组7只未种植细胞的人工血管分别移植犬颈总动脉。移植2个月后,观察两组移植人工血管的通畅率。结果种植到人工血管表面的内皮祖细胞能持续分泌t-PA和NO等抗血栓物质。行置换术后2个月,实验组人工血管通畅率高于对照组,扫描电镜见实验组人工血管腔内形成完整融合的内皮细胞单层。结论内皮祖细胞种植能加快人工血管内皮化和增强其抗血栓能力,有效改善小径人工血管的在体移植通畅率。
Objective To investigate the effect of endothelial progenitor ceil(EPCs) seeding on transplantation patency of small diameter polyurethane artificial vessels, in order to find a novel strategy for improving the patency of small diameter artificial vessel after transplantation. Methods The peripheral blood mononuclear ceils of healthy adult were induced to form EPCs. When EPCs were seeded on the luminal surface of small diameter artificial vessels, the secretion of tissue-type plasminogen activator (t-PA) and nitric oxide (NO) by EPCs were measured by enzyme-linked immunosorbent assay and nitrate reductase methods. Twelve domestic dogs were randomly divided into two groups. The canine carotid arteries of experiment group were transplanted with EPCs-lined small diameter polyurethane artificial vessels ( n = 6), while those of control group were transplanted with unseeded ones ( n = 7). The transplantation patency of small diameter polyurethane artificial vessels in two groups was observed two months after operation. Results Two months after the operation, the patency rate of small diameter artificial vessels of experiment group was higher than that of control group. Scanning electron microscopic observation showed that the luminal surfaces of the grafts of experiment group two months after implantation were covered with a confluent monolayer of endothelial ceils. Conclusion Seeding EPCs can promote endothelization of mall diameter artificial vessels and enhance their antithrombotic potential, which significantly improve the patency rate of small diameter artificial vessels in vivo.
