目的对比脂质体与电穿孔法及不同载体对大鼠雪旺细胞的转染效率,为进一步研究神经胶质细胞奠定实验基础。方法以大鼠雪旺细胞为研究对象,采用脂质体法和电穿孔法分别转染绿色荧光蛋白(GFP)载体和羧基荧光素(FAM)标记的miRNAmimics,比较和分析两者的转染效率。结果脂质体转染法成功转染GFP质粒大鼠雪旺细胞的效率为(15.6±2.3)%,转染miRNAmimics的效率为(36.8±3.5)%;采用电穿孔法转染GFP质粒的效率高达(40.6±3.3)%,而该法转染miRNAmimics的效率则非常低。结论大鼠雪旺细胞的转染效率可能与转染方式、被转染物的大小和形式有关。
Objective To compare the transfection efficiency between different transfection methods and different vectors in rat Schwann cells so as to provide experimental basis for further study of glial cells. Methods GFP plasmid and FAM-labeled miRNA mimics were transfected into rat Schwann cells by using lipofection and electroporation methods, respectively. And the transfection efficiency of the two methods was compared. Results The efficiency of GFP vector and miRNA minmics transfected by lipofection was (15.6 ± 2.3) % and (36.8±3.5)%, respectively. Compared with lipofection method, the efficiency of GFP vector transfected by electroporation was up to (40.6±3.3)%, while the efficiency of miRNA minmics was rather low. Conclusion The efficiency of rat Schwann cells transfection is probably associated with transfection method as well as the size and form of the transfected complexes.
