目的探讨3'-大豆苷元磺酸钠抗谷氨酸引起的PC12细胞损伤的影响。方法采用谷氨酸诱导PC12细胞损伤模型,分别给予3.0、10.0μmol/ml的3'-大豆苷元磺酸钠干预,采用MTT法检测PC12细胞活力;同时测定培养液中乳酸脱氢酶活性、超氧化物歧化酶活性和丙二醛的含量。结果3.0、10.0μmol/ml的3'-大豆苷元磺酸钠能提高谷氨酸引起的PC12细胞的存活率;3.0、10.0μmol/ml的3'-大豆苷元磺酸钠能抑制由谷氨酸引起的乳酸脱氢酶活性;3.0、10.0μmol/ml的3'-大豆苷元磺酸钠能提高超氧化物歧化酶活性和降低丙二醛含量。结论3'-大豆苷元磺酸钠对谷氨酸引起的PC12细胞损伤具有明显的保护作用,其保护作用可能与抑制由谷氨酸引起的乳酸脱氢酶活性,提高超氧化物歧化酶活性和降低丙二醛含量有关。
Objective To study the anti-glutamate induced PC12 cell damage effect of 3'-daidzein suffocates sodium (3 '-DSS). Methods The PC12 cell damage models went induced by glutamate,different doses of 3'-DSS(3.0,10.0μmol/ml) were sub-jected to low,moderate and high dose treating groups respectively. The PC12 cell viability was tested by-MTT,the activity of lac-tate dehydrogenase,the contents of SOD and MDA were also tested. Results The PC12 cell survival rate in all treating groups is higher than it in the control-group,all the three doses of 3'-DSS can inhibit the lactate dehydrogenase activity,3.0 and 10.0μmol/ml of 3'-DSS can increase the SOD activity and decrease the content of MDA. Conclusion 3'-DSS show significant protect ef-fect of PC12 cell damage induced by glutamate. The mechanism may be inhibiting the lactate dehydrogenase activity ,increasing the SOD activity and decreasing the MDA content.
