目的为研究miR375对肌侵蛋白(MTPN)表达的影响,探寻miR375在糖尿病中的作用。方法该研究用质粒pAAV-miR375转染Nit-1细胞48h后,分别通过Real time RT-PCR和Northern Blot检测Nit-1细胞中miR375的表达,Western Blot检测Nit-1细胞中MTPN蛋白的表达。结果在转染pAAV-miR375的Nit-1细胞中miR375的表达明显升高,是转染空质粒pAAV-MCS和未转染的6倍。MTPN蛋白的表达与转染了空质粒pAAV-MCS和未转染的Nit-1细胞相比,明显下降。结论在Nit-1细胞中miR375过表达引起了MTPN蛋白表达下调,推测miR-375可能是通过调节MTPN基因的表达来调控胰岛素分泌的,从而影响体内血糖的浓度。
[ Objective ] To study the effect on the expression of Myotrophin (MTPN) and finding the function of miR375 in diabetes. [ Methods ] 48 hour after the pAAV-miR375 plasmid transfeeted into the Nit-1 cells, the expression of miR375 in the Nit-1 cells was analyzed by Real-time PCR and Northern Blot. The protein expression of MTPN in the Nit-1 cells was analyzed by western Blot. [ Results ] The expression of miR375 was increase 6 times in the Nit-1 cells transfected with pAAV-miR375, camper to the Nit-1 cells transfeeted with control plasmid or untransfected. The expression of MTPN was reduced apparently in the Nit-1 cells transfected with pAAV-miR375, camper to the Nit-1 cells transfected with control plasmid or untransfeeted. [ Conclusion ] Over-expression of miR- 375 reduced the expression of Mtpn in Nit-1 cells in vitro, adjusting the blood glucose.
