从蛙虹彩病毒(Rana grylio virus,RGV)基因组中克隆了含凋亡相关结构域的新基因-cop ( Caspase recruitment domain only protein, COP)基因的全部编码区,成功构建了重组表达载体,进行了原核表达,并在鲤鱼上皮瘤细胞 (Epithelioma papulosum cyprini , EPC)中进行了亚细胞定位.序列分析表明,RGV cop基因全长288bp,编码一个长为95aa,分子量为10.4×10^3的推定蛋白.二级结构预测表明其含有5个α螺旋.同源性比对分析显示,RGV cop与其他6株虹彩病毒及人类cop相关蛋白同源性最高为94%,最低为33%.重组原核表达质粒pET28a—COP转化大肠杆菌BL21后,经IPTG诱导表达,获得一条约14×10^3的融合蛋白.重组真核表达质粒pEGFPN3-COP转染EPC细胞,经DAPI染色后,在荧光显微镜下观察显示重组蛋白在整个细胞中均有分布.
Cop(Caspase recruitment domain only protein,COP) gene which contains apoptotic domain of Rana grylio virus (RGV COP) was cloned, sequenced and expressed in E. coli and Epitheliorna papulosurn cyprini (EPC) cells. From the results of sequence analysis, the cop gene is 288 bp long and encodes a protein of 95 aa with a predicted molecular mass of 10.4× 10^3. Alignment of the deduced amino acid sequence of RGV COP with Homo sapiens caspase-1 dominant-negative inhibitor pseudo-ICE isoform 2 (Human COP) and other six iridovirus showed that they had the homology degree of 33% to 94%. The recombinant prokaryotic and eukaryotic expression plasmid containing this fragment was constructed. The former expressed in E. coll. BL21 and an expected 14 ×10^3 fusion protein was isolated by SDS-PAGE. EPC cells transfected with the later plasmid were stained with DAPI and observed under a Leica DM IRB fluorescence microscope. Green fluorescence was observed in whole cell. It's first report of this gene in iridovirus.
