目的探讨中药黄芪注射液(AI)对微囊藻毒素LR(MCLR)所致大鼠海马急性损伤的化学预防作用及机制。方法采用海马内注射MCLR制作毒素急性损伤模型,通过组织病变的病理学观察和生化检测超氧化物歧化酶(SOD)、过氧化氢酶(CAT)、谷胱甘肽过氧化物酶(GPx)活性及丙二醛(MDA)含量的改变等实验方法观察黄芪注射液的作用。结果与对照组相比较,MCLR使海马CA1区细胞排列不规则,水肿、核固缩,海马MDA含量增加(P〈0.05),抗氧化酶SOD、CAT、GPx活性增加(P〈0.05),黄芪干预后能减轻海马CA1区形态学损伤,降低MDA含量(P〈0.05),抗氧化酶SOD、CAT、GPx活性明显下降(P〈0.05)。结论黄芪注射液可改善MCLR导致的大鼠海马CA1区的病理改变,减少海马部位的脂质过氧化。
Objective In order to investigate the chemopreventive effects of Astragalus injection on acute hippocampal injury by microcysitn-LR of rats.Methods To investigate the chemopreventive effects of Astragalus injection on acute hippocampal injury induced by microcysitn-LR in rats.Results Intrahippocampal administration of MCLR caused pathological changes of neuron injury in hippocampal CA1 area.Pyramidal cellular nuclear was shrinked and accompanied by cellular edema.Furthermore,the pyramidal layer was disordered and irregular in the arrangement.Compared with control group,the MDA content and the activities of SOD,CAT,GPx were enhanced in model group(P0.05).Compared with model group,the abnormal morphology in hippocampal CA1 area and the increase of MDA level and antioxidant enzyme activities were significantly relieved by intraperitoneal administration of Astragalus injection(P0.05).Conclusion Astragalus injection can inhibit lipid peroxidation and relieve the abnormal morphology induced by MCLR in hippocampal CA1 aera.
