目的:观察槐米对S180荷瘤小鼠的抑瘤作用及作用机制。方法:采用MTT法测定在槐米干预下,S180细胞在24,48,72h的增殖活性。将S180细胞皮下注射接种于小鼠左前肢腋下建立荷瘤小鼠模型,设正常组、模型对照组、槐米组(50、100、200mg/kg)及环磷酰胺组(25mg/kg)。造模后连续灌胃给药4 w,1次/d。测各组小鼠体重及瘤组织、脾脏、胸腺重量,并计算抑瘤率、胸腺系数及脾系数。比色法测定血清中超氧化物歧化酶(SOD)、丙二醛(MDA)水平,ELISA法检测血清中白细胞介素-2(IL-2)、肿瘤坏死因子-α(TNF-α)、血管内皮生长因子(VEGF)、碱性成纤维细胞生长因子(b FGF)、基质金属蛋白酶-9(MMP-9)水平。结果:槐米在31.25-500μg/ml范围内对体外培养48h的S180细胞具有明显抑制作用,呈剂量依赖性。与模型对照组比较,槐米提取物在50-200mg/kg剂量范围内可不同程度增加小鼠体重、胸腺系数、脾系数,抑瘤率最高可达29%;升高小鼠血清中SOD、IL-2水平,降低MDA、TNF-α、VEGF、b FGF、MMP-9水平,且在100mg/kg剂量下作用最明显。结论:槐米对S180荷瘤小鼠肿瘤具有抑制作用,其机制与增加血清中SOD、IL-2水平,降低MDA、TNF-α、VEGF、b FGF、MMP-9水平相关。
Objective: To observe inhibition effects and analyze its mechanisms of sophoraflower bud abstract on tumor in S180 tumor-bearing mice.Methods: The proliferative activities of S180 cells at 24,48,72 h under sophora flower bud abstractinterfering were determined by MTT method. Set up tumor-bearing mice models by injecting subcutaneously S180 cells into armpit of left forelimb in mice. The mice were randomly divided into normal group,model control group,sophora flower bud abstract groups with different doses( 50,100,200 mg / kg) and cyclophosphamide group( 25 mg / kg). The mice were given by ig. for 4 w,onceper day. The levels of superoxide dismutase( SOD),methane dicarboxylic aldehyde( MDA) were detected by colorimetric method and the level of IL-2,TNF-α,vascular endothelial growth factor( VEGF),basic fibroblast growth factor( b FGF),matrix metalloproteinase( MMP-9) in serum were determined by ELISA. Measure weight of body,tumor,spleen,thymus of mice,and calculate tumor inhibition rate,thymus coefficient and spleen coefficient. Results: Sophora flower bud abstract at a dose-range 31. 25 ~ 500μg / ml could inhibit dose-dependently S180 cells cultured for 48 h. Compared with the model control group,sophora flower bud abstract at a dose-range 50 ~ 200 mg / kg could increase body weight,thymus coefficient and spleen coefficient,and tumor inhibition rate could top 29%; and increase the levels of SOD,IL-2,and decline the levels of MDA,TNF-α,VEGF,b FGF,MMP-9 in serum( P 〈 0. 05,P 〈 0. 01). Conclusion: Sophora flower bud abstract had inhibition effects on tumor in S180 tumor-bearing mice,which may be related to increase the levels of SOD,IL-2,and decline the levels of MDA,TNF-α,VEGF,b FGF,MMP-9 in serum in mice.
