目的:在裸鼠体内实验中,观察使用人端粒酶逆转录酶(hTERT)核心启动子引导人钠碘转运体(hNIS)基因的重组腺病毒介导放射性核素显像的可行性。方法:扩增重组腺病毒,测定转染后U87细胞摄碘率,构建U87人胶质瘤荷瘤裸鼠模型,使用重组腺病毒Ad—hTERT—hNIS进行体内转染,进行放射性核素^99Tcm显像。结果:重组腺病毒Ad—hTERT—hNIS转染端粒酶阳性的U87细胞后,hNIS基因能够介导胶质瘤细胞摄碘,并且该病毒能对U87胶质瘤荷瘤裸鼠进行体内转染;成功介导靶向性hNIS基因的表达,而转染后的荷瘤裸鼠模型能成功介导放射性核素9STcmO2^-显像。结论:重组腺病毒Ad—hTERT—hNIS能够实现裸鼠体内hNIS基因的靶向性表达,并能介导放射性核素99Tcm显像。
Objective: To investigate the possibility of transfecting human sodium iodide symporter (hNIS) gene following human telom- erase reverse transcriptase promoter (hTPO) into U87 malignant glioma nude mice for radioactive nuclide imaging in vivo. Methods: The recombinant adenovirus were amplified and purified, and the ^125I-iodide uptake of transfected U87 cells was detected. The malignant glioma nude mice animal model was performed, Ad-hTERT-hNIS was transfected in vivo, then radioactive nuclide ^99Tcm tumor imaging was obtained. Results: After U87 cells of telomerase-positive were transfected with Ad-hTERT-hNIS, Ad-hTERT-hNIS could mediate ^125I uptake of malignant glioma cell and malignant glioma nude mice animal model could be transfected by Ad-hTERT-hNIS, result in tumor specific hNIS gene expressioning and radioactive nuclide ^99Tcm tumor imaging. Conclusion: The recombinant adenovirus Ad- hTERT-hNIS could lead tumor specific hNIS gene expression in malignant glioma nude mice animal model and show ^99Tcm tumor imaging.
