目的研究灵猫方抗肝纤维化的作用及其机制。方法55只BALB/c小鼠随机分为4组:正常组、模型组、秋水仙碱组、灵猫方组。ConA(15mg/kg)尾静脉注射制作肝纤维化模型,每周两次,共10周。正常组给予双蒸水灌胃,其余组分别给予秋水仙碱(0.2ms/ks)、灵猫方(7.5g/kg)。末次灌胃24h后处死小鼠,检测肝脏病理变化以及血清ALT、AST、ALB、DBIL表达水平、肝组织羟脯氨酸(HYP)含量变化、肝组织纤维化蛋白(FBRS)的表达水平。结果(1)与模型组相比,灵猫方组血清ALB水平明显增加,DBIL水平明显降低(P均〈0.01),HYP的含量明显降低(P〈0.05)。(2)肝脏病理学检测,灵猫方组肝纤维化的程度和胶原显色指数较模型组明显减轻(P〈0.05,P〈0.01)。(3)与正常组相比,模型组的FBRS表达水平明显增加(P〈0.05),与模型组相比,灵猫方组肝组织中FBRS的表达水平明显降低(P〈0.05)。结论中药灵猫方通过降低肝组织纤维化蛋白的表达发挥抗肝纤维化的作用。
Objective To investigate the mechanism of anti-hepatic fibrosis of Lingmao formula. Methods 55 BALB/c mice were divided into 4 groups, namely the normal group, the model group, the positive control group and the treatment group. The hepatic fibrosis model was established by tail vein injection of ConA( 15 mg/kg), twice a week, for 10 weeks. The normal and the model groups were administered ddH20 with dose 10 ml/kg,and the positive control group were administered colchicine (0. 2 mg/kg), and the treatment group were administered Lingmao formula (7. 5 g/kg ). Twenty-four hours after the last gastric perfusion, the mice were sacrificed. The serum and tissue samples were collected to detect histopathological changes and the expression levels of ALT,AST, ALB and DBIL in the serum, and the expression level of hydroxyproline (HYP) in the liver. The expression level of fibrosin (FBRS) was examined. Results ( 1 ) Compared with model group, ALB level was increased and DBIL level was decreased ( P 〈 0. 01 ), and HYP level was declined ( P 〈 0. 05 ) in Lingrnao Formula group. (2)Compared with the model group,the degree of hepatic fibrosis and the content of collagen were decreased (P 〈 0. 05 ,P 〈 0. 01 ). (3)Compared with the normal group, the expression level of FBRS in the model group was increased significantly(P 〈 0. 01 ), and compared with the model group, the expression level of FBRS in Lingmao formula group was decreased( P 〈 0. 05 ). Conclusions The mechanism of anti-hepatic fibrosis of Lingrnao formula is through decreasing the expression of FBRS.
