目的观察创伤后应激障碍(PTSD)样行为异常大鼠蓝斑神经元缝隙连接蛋白43(Connexin43,Cx43)表达变化,探讨PTSD的发病机制。方法采用国际认定的SPS方法刺激建立大鼠PTSD模型,取成年健康雄性Wistar大鼠100只,随机分为连续单一刺激(single prolonged stress,SPS)模型1d、4d、7d、14d组和对照组,应用免疫组化、免疫印记和逆转录-聚合酶链式反应(RT-PCR)方法检测PTSD大鼠蓝斑神经元缝隙连接蛋白43的表达变化。结果经SPS刺激后大鼠蓝斑神经元细胞内Cx43蛋白和mRNA于1d开始逐渐升高,4d时表达最多,之后逐渐下降。结论创伤后应激障碍模型大鼠蓝斑Cx43的表达变化,可能与PTSD大鼠蓝斑功能异常相关。
Objective To observe the changes of connexin43(Cx43) in the locus coeruleus neurons of posttraumatic stress disorder(PTSD) rats to explore the pathogensis of PTSD.Methods The single prolonged stress(SPS) method was used to set up the rat PTSD models.A total of 100 healthy male Wistar rats were randomly divided into 1dSPS,4dSPS,7d SPSand 14d SPS groups and a normal control group.Immunohistochemistry,western blot and RT-PCR were used to detect the changes of the expressions of Cx43 in the locus coeruleus neurons. Results The expression levels of Cx43 protein and mRNA in the locus coeruleus gradually increased at 1d after SPS,and reached the maximum at SPS 4d,and then gradually decreased.Conclusion The changes of Cx43 expression in t he locus coeruleus of PTSD rat may be related to the pathogenesis of PTSD rats.
