目的在人食管鳞状细胞癌组织和癌旁正常黏膜组织中检测细胞外信号调节激酶5(ERK5)蛋白和mRNA表达情况,对照临床病理参数,探讨ERIC5表达的临床意义。方法选取食管鳞癌组织(食管鳞癌组)和癌旁正常组织(正常组)各115例,采用免疫组化法检测两组ERK5蛋白表达,RT—PCR法检测ERK5mRNA相对表达量,并分析ERK5蛋白、mRNA表达与食管鳞癌临床病理参数的关系。结果食管鳞癌组及正常组ERK5蛋白表达阳性率分别为90.4%和68.6%,两组ERK5蛋白表达有显著差异(P〈0.05);食管鳞癌组及正常组ERK5mRNA的相对表达量分别为O.82±0.07、0.61±0.02,两组ERK5mRNA表达有显著差异(P〈0.05)。不同的病理分级分期的食管鳞癌ERK5蛋白、mRNA表达有显著差异(P〈0.05)。而不同的性别、年龄的食管鳞癌ERK5蛋白、mRNA表达无显著差异(P〉0.05)。ERK5蛋白和mRNA表达呈正相关。结论ERK5蛋白和mRNA表达在癌组织中较癌旁正常组织高;分期越高分化程度越低及发生淋巴结转移的食管鳞癌组织中ERK5蛋白和mRNA的表达水平越高。提示高表达的ERK5和食管癌的恶性程度和转移密切相关。
Objective To detect the expression of extracellular signal regulated kinase 5 ( ERK5 ) protein and mRNA in human esophageal squamous cell carcinoma tissue and adjacent normal mucosa and to analyze the clinical and pathological parameters and the clinical significance of ERK5 expression. Methods 115 cases of esophageal squamous cell carcinoma( ESCC group)and normal tissues (normal group)were selected and the immunochemical method was used to to detect ERK5 protein expression. The quantity of ERK5 mRNA relative expression was detec- ted by RT- PCR method, and the relationship between ERK5, mRNA protein expression and clinical pathological parameters of esophageal squamous cell carcinoma were analyzed. Results the positive rate of ERK5 protein expres- sion were 90.4% and 68.6% in esop shown significant different ( P 〈 0.05 tively in esophageal squamous cell hageal squamous cell carcinoma group and the normal group, respectively and ) ;the amount of ERK5 mRNA were 0.82 ±0.07 and 0.61 ±0.02,respec- carcinoma group and the normal group and shown significant difference ( P 〈 0.05 ). There were significant differences in different pathological grading of ERK5 protein and mRNA expres- sion in different pathological stage of esophageal squamous cell carcinoma(P 〈 0.05 ) ;but there had no significant difference(P 〉 O. 05 )in different gender, age of esophageal squamous cell carcinoma ERK5 protein and mRNA ex- pression. ERK5 protein and mRNA expression showed positive correlation. Conclusion Compared with adjacent nor- mal tissues, ERK5 protein and mRNA expression in cancerous tissues are shown high expression level;the higher the pathological stages or less the differentiation degree is, the lymph node metastasis, ERK5 protein and mRNA ex- pression level is in the squamous cell carcinoma of the esophagus, suggesting that higher level expression of ERK5 is related with the degree of malignant and metastasis in esophageal cancer.
