Fraxinus hupehensi is a tall deciduous tree of OleaceaeFraxinus originating from Hubei Jingshan and Zhongxiang, and belongs to national rare and endangered plants. TakingFraxinus stems as explants, this study explored the sterilization time and method and the best conditions for callus induction. Results show that the sterilization effect is the best when tender stem is treated by alcohol （75%） for 30 s and HgCl2for 10 min. The best formula for callus induction is MS＋BA 2.0 mg/L＋2, 4-D 1.5 mg/L.
