目的建立家兔耳部VX-2肿瘤模型,探讨磁感应加温方法对肿瘤的杀伤效应。方法于家兔耳部皮下注射VX-2肿瘤细胞悬液建立兔耳肿瘤模型,局部注射纳米Fe3O4微粒磁液并进行磁感应加温,观察瘤体大小及组织病理变化。结果兔耳皮下接种2×10^6个瘤细胞10天后能形成长径〉5mm的类圆形VX-2肿瘤,利用此模型可大体观察磁液在肿瘤局部的聚集情况;400mg/ml的磁液50—100μl局部注射能在15kA/m的交变磁场中使肿瘤组织有效升温至43℃以上,并发生坏死。结论家兔耳部肿瘤模型易于建立和观察,利用该模型证明磁感应加温能对瘤组织产生明显的杀伤作用。
Objective To establish an model of VX-2 tumor on rabbit ears and discuss the anti-tumor effect of magneticinduction hyperthermia. Methods The model was established by injecting VX-2 cell suspension subcutaneously into the rabbit ears. Magnetic-induction hyperthermia was then performed on the tumor areas after local injection of Fe3O4 magnetic fluids. The volume and pathology of the tumor were observed. Results Round-llke VX-2 tumor sized over 5 mm in long diameter was observed ten days after injection of 2 × 10^6 tumor cells into the rabbit ears subcutaneously. Aggregation of magnetic fluids in the tumor areas could be observed macroscopically in the model. The temperature of the tumor tissues was increased by more than 43 ℃ in 15 kA/m alternating magnetic field after local injection or50 - 100 μl magnetic fluids (400 mg/ml) . Necrosis could be observed in the tissues. Conclusions As a convenient tool for microscopical observation, the VX-2 tumor model can be established easily in rabbit ears. Anti-tumor effect of magnetic-induction hyperthermia is demonstrated in this model.
