目的:观察外源性一氧化碳释放分子(carbon monoxide releasing-moleculesⅡ,CORM-2)对盲肠结扎和穿孔(cecal ligation and puncture,CLP)脓毒症模型小鼠血小板膜糖蛋白CD41、CD61和CD62p表达的影响。方法:C57BL/6小鼠60只,随机均分成4个组,假手术组、CLP组、CLP+CORM-2组和高浓度氧预处理+CLP+CORM-2组,每组各15只,每组再分6 h、12 h和24 h 3个时间段。用流式细胞仪测定血小板膜上CD41、CD61的平均荧光强度及CD62p的阳性率。结果:各时间段高氧预处理联合CORM-2干预组血小板膜糖蛋白CD61和CD62p的表达较CLP组明显下降,接近于假手术组水平。结论:CORM-2可降低CLP小鼠血小板膜糖蛋白CD61和CD62p的表达,抑制血小板的活化。
Objective: To investigate the effects of extrinsic carbon monoxide-releasing moleculesⅡ(CORM-2) on the expressions of CD41/CD61 and CD62p on the platelet membrane surface in mice with cecal ligation and puncture(CLP).Methods: Sixty male C57BL/6 mice were randomly divided into sham,CLP,CLP+CORM-2,O2+CLP+CORM-2 groups.One group were randomly divided into 6 h,12 h and 24 h.The mean immunofluorescence intensity of CD41,CD61or positive percentage of CD62p was measured by the flow cytometry(FCM).Results: The mean fluorescence intensity of CD61 and positive percentage of CD62p on platelet membrane respectively in O2+CORM-2+CLP group were significantly decreased while compared with CLP group and were approximated with control Sham group.Conclusion: The extrinsic carbon monoxide-releasing moleculesⅡ may induce down regulation of platelet membrane glycoprotein CD61 and CD62p expression in CLP mice,with inhibiting that platelet activation.
