目的研究氧化型胆固醇之一3β,5α,6β-三羟胆固醇(Triol)对生物膜结构的影响。方法以人红细胞膜为体外生物膜模型,采用原子力显微镜(AFM)和傅里叶变换红外光谱(FTIR)分析Triol作用下红细胞膜结构的变化。结果AFM观察到Triol使红细胞膜表面对应膜蛋白的突起变得平缓。FTIR分析显示Triol使红细胞膜结构,尤其是膜蛋白二级结构发生明显改变,主要表现为一定浓度Triol使膜蛋白中α-螺旋和β-转角含量显著增加,β-折叠含量明显减少。结论氧化型胆固醇插入生物膜后改变膜结构,这可能是其导致细胞凋亡的机制之一。
Objective To investigate the effect of oxysterol cholestane-3β, 5α, 6β-triol (Triol) on the structure of biological membrane. Methods Human erythrocyte membrane was used as a biological membrane model in vitro. The structure of erythrocyte membrane was analyzed by atomic force microscopy (AFM) and Fourier transform infrared spectroscopy (FTIR). Results As observed by AFM, on the surface of erythrocyte membrane treated with Triol, the protusions, which indicated membrane proteins, were much flatter than control group. FTIR study revealed that Triol significantly changed the structure of erythrocyte membrane, especially the secondary structure of membrane proteins. Triol at indicated concentrations significantly increased α-helix and β-turn, and decreased β-sheet. Conclusion The alteration of structure of biological membrane inserted oxysterols might be one of mechanisms by which oxysterols induced cell apoptosis.
