目的:研究鲫鱼卵唾液酸糖肽(Ca—SGPP)对成骨细胞MC3T3-E1骨形成活性及OPG/RANKL基因表达水平的影响。方法:在MC3T3-E1细胞中加入不同浓度的Ca—SGPP,采用M1-r法、试剂盒和茜素红染色法检测Ca—SGPP对MC3T3—E1细胞增殖、分化和矿化的影响;刺用逆转录聚合酶链式反应(1it—PCR)技术检测MC3T3-E1细胞骨形态发生蛋白-2(BMP-2)、骨保护素(OPG)及核因子KB受体活化因子配体(RANKL)基因mRNA表达水平。结果:Ca—SGPP可显著促进MC3T3-E1细胞增殖及ALP、OCN和COLI的分泌,提高MC3T3-E1细胞矿化能力。上调其BMP一2表达水平及OPG/RANKL的比值,且分子质量大的糖肽效果更佳。结论:Ca—SGPP具有促进MC3T3-E1细胞增殖、分化和矿化的能力,其机制可能与OPG/RANKL比值上调有关。
Objective: To investigate the effects of sialyglycopeptides from the eggs of Carassius auratus(Ca-SGPP)on the proliferation, diferentiation, mineralization and the gene expression of OPG/RANKL in MC3T3-E1 cells. Method:MTT assay, alkaline phosphatase(ALP) activity assay and calcium deposition were determined, the secretion of collagen I(COL I) and osteocalein(OCN) were assayed by enzyme-linked immunosorbent assay(ELISA), the m RNA expression levels of bone morphogenetic protein 2(BMP-2), osteoprotegerin(OPG) and receptor activator for nuclear factor-κB ligand(RANKL) were also measured by reverse transcription polymerase chain reaction(RT-PCR). Results: The Ca-SGPP could effectively promote the proliferation(P〈0.01), increase ALP,OCN and COL I content, enhance the mineralization ability, improve the expression levels of BMP-2 and OPG/RANKL ratio of MC3T3-E1 cells. Furthermore, the effect of Ca-SGPP of more than 10 ku was much better than that of less than 10 ku. Conclusion: The Ca-SGPP can remarkably promote the osteogenic activity on MC3T3-E1 cells, and its mechanism is probably related to the rise of OPG/RANKL rato.
