目的:观察大鼠脑、小肠肌间神经丛神经元和血浆内胃动素的表达.探讨胃动素在水浸加束缚应激性胃溃疡中的作用。 方法:实验于2004—10/2005—10在青岛大学医学院完成。选择健康雄性Wister大鼠76只,随机分为水浸加束缚组10只;侧脑室注射胃动素组32只;皮下注射N-硝基精氨酸酯+侧脑室注射胃动素组8只和相应的3个对照组:正常对照组10只;侧脑室注射生理盐水组8只;皮下注射N-硝基精氨酸酯+侧脑室注射生理盐水组8只。采用放射免疫分析、荧光免疫组化双染和神经生理学等实验方法,观察脑、小肠肌间神经丛和血浆内胃动素的表达,探讨胃动素对大鼠束缚加水浸诱导的应激性胃溃疡的影响。 结果:76只大鼠均进入结果分析。①在正常大鼠小肠肌间神经丛内和原代培养的肠肌间神经结神经元均可见有胃动素样免疫反应阳性物的表达,且胃动素与一氧化氮合酶共同表达于同一神经元内,但胃动素不与消化道感觉性神经元内特有的钙结合蛋白共存。②在大鼠应激性胃溃疡产生的同时,其血浆内胃动素的含量明显少于正常对照组[(162.86±25.25),(221.54±31.82)ng/L,P〈0.05],而下丘脑、延脑、垂体和肌间神经丛神经元内胃动素的含量明显高于正常对照组[(127.66±9.61),(74.34±19.63)ng/g;(40.56±3.17),(17.26±6.55)ng/g;(28.97±3.24),(11.42±1.25)ng/g;(27.97±2.20),(13.81±1.05)ng/g,P〈0.05~0.01]。③侧脑室注射胃动素大鼠应激性胃溃疡的产生明显减少,且呈明显的量效依赖关系,(P〈0.05~0.01),但经皮下注射一氧化氮合酶抑制剂N-硝基精氨酸酯后,脑室注射胃动素,胃动素的胃黏膜细胞保护作用消失(溃疡数:89.00±12.61.81.12±11.39.P〉0.05)。 结论:胃动素与一氧化氮?
AIM: To observe the expressions of motilin in brain, neuron of intestinal myenteric plexus and plasma, and probe into the effect of motilin on water immersion and stress induced gastric ulcer. METHODS: The experiment was performed in the Medical College of Qingdao University from October 2004 to October 2005. Seventy-six healthy male Wistar rats were randomly divided into water immersion restraint group (n=10), intracerebroventricular (icv) injection of motilin group (n =32), subcutaneous (sc) Nto-Nitro-L-arginine methylester (L-NAME)+ icv injection of motilin group (n=8) and three corresponding control groups: Normal control group (n=10); Icv injection of normal saline group (n=8); Sc L-NAME+ icv injection of normal saline group (n=8). The expressions of motilin in brain, myenteric plexus and plasma were observed and detected with radio-immunoassay (RIA) and fluor-immunohistochemical double staining etc. Probed into the effect of motilin on water immersion and stress induced gastric ulcer in rats. RESULTS:A total of 76 rats were involved in the analysis of results. ① Expression of immunoreactive positive neurons of motilin could be seen in small intestinal myenteric plexus and primarily cultured intestinal ganglia neurons, there were expressions of both motilin and nitric oxide synthase (nNOS) in the same neuron, while the motilin didn't coexisted with special calcium binding protein in the sensitive neurons of alimentary canal. ②At the same time of water immersion and stress induced gastric ulcer in rats, the content of motilin in plasma was significantly lower than that in the normal control group [(221.54±31.82), (162.86±25.25) ng/L,P 〈 0.05], while the contents of motilin in neurons of hypothalamus, medulla oblongata, pituitary and myenteric plexus were markedly higher than those in the normal control group [(74.34±19.63), (127.66±9.61) ng/g; (17.26±6.55), (40.56±3.17) ng/g; (11.42±1.25), (28.97±.24) ng
